methylation status of p15 gene in esophageal squamous cell carcinoma using methylation-specific pcr

چکیده

introduction: promoter methylation has an inhibitory effect on gene expression. thus we investigated methylation patterns of p15 gene in esophageal squamous cell carcinoma (escc), one of the most frequent and fetal cancers in the world. material and methods: p15 promoter methylation status of 30 escc and 10 normal specimens was investigated using methylation specific pcr. dna extracted from the specimens was digested by restriction enzyme. the digested dna was modified by sodium bisulfite. using methylated and unmethylated specific primers, methylation status was determined based on the presence of the methylated and unmethylated specific pcr products. also dna extracted from normal blood and cpg methylase (sssi)–treated genomic dna were used as the negative and positive controls respectively. results: our results showed that all normal specimens were unmethyalted and only 5 out of 30 tumor samples (16.6%) were methylated. statistical analysis (fisher’s exact tests) showed no significant correlation between p15 gene promoter methylation and escc cancer. conclusion: according to the results of the study, p15 gene does not play a significant role in aberrant methylation process in escc, other mechanisms may involve in p15 inactivation during escc development. however, due to limited number of specimens, we recommend further investigation using a larger sample.

Analysis of SEPT9 Gene Promoter Methylation Status in Esophageal Squamous Cell Carcinoma

Introduction: The changes in the level of SEPT9 gene promoter methylation can contribute to the formation of esophageal squamous cell carcinoma. The aim of this study was to evaluate the level of changes in the level of SEPT9 gene promoter methylation in the esophageal squamous cell carcinoma. Methods: In the present case-control study, we collected 75 paraffin blocks of esophageal cancer tiss...

Methylation and expression of PTPN22 in esophageal squamous cell carcinoma

Esophageal squamous cell carcinoma (ESCC) is a fatal disease contributed by both genetic and epigenetic factors. The epigenetic alteration of protein tyrosine phosphatase non-receptor type 22 (PTPN22) and its clinical significance in ESCC were still not yet clarified. A quantitative methylation study of PTPN22 and its expression were conducted in 121 and 31 paired tumor and adjacent normal tiss...

PRSS8 methylation and its significance in esophageal squamous cell carcinoma

Esophageal cancer is one of the most common cancers worldwide, and the incidence and mortality is increasing rapidly in recent years in China, but the underlying mechanisms are largely unclear. Herein we found that the expression of PRSS8, a serine protease prostasin, is significantly decreased in esophageal squamous cell carcinomas (ESCC) at mRNA and protein levels. The reduction of PRSS8 was ...

Two Steps Methylation Specific PCR for Assessment of APC Promoter Methylation in Gastric Adenocarcinoma

Gastric Cancer (GC) is the second most common cancer in the world and a leading cause of cancer-related mortality. Methylation of promoter CpG islands (CGIs) belonging to tumor suppressor genes causes transcriptional silencing of their corresponding genes leading to carcinogenesis and other disorders. Adenomatous Polyposis Coli (APC) a tumor suppressor gene is inactivated by methylation of prom...

DNA Methylation Status of PAX1 and ZNF582 in Esophageal Squamous Cell Carcinoma

Hypermethylation of specific gene promoters is an important mechanism of carcinogenesis. A high frequency of promoter methylation of PAX1 and ZNF582 genes has been detected in cervical cancer. In the present study, we investigated the methylation status of PAX1 and ZNF582 genes in esophageal squamous cell carcinoma (ESCC) tissues. Tumor and paracancerous tissues were obtained from 14 ESCC patie...

Prognostic CpG Methylation Biomarkers Identified by Methylation Array in Esophageal Squamous Cell Carcinoma Patients

BACKGROUND Esophageal squamous cell carcinoma (ESCC) is an aggressive cancer with poor prognosis. We aimed to identify a panel of CpG methylation biomarkers for prognosis prediction of ESCC patients. METHODS Illumina's GoldenGate methylation array, supervised principal components, Kaplan-Meier survival analyses and Cox regression model were conducted on dissected tumor tissues from a training...
